Spontaneous oxidative modification of blood plasma proteins in patients of reproductive age with cataract

Purpose:

study the spontaneous oxidative modification of blood plasma proteins in patients of reproductive age with cataracts.

Setting:

Department of ophthalmology, Crimea State Medical University named after S.I.Georgievsky, Simferopol, Crimea

Methods:

Were examined 60 patients with senile cataract (SC) and no associated ocular disease, aged 38 to 55 years, mean age 42,2 ± 4,31 years, males and females were equal. The control group (CG) -34 healthy persons, comparable in sex and age, with the group AC. Smoking, diabetes mellitus, inflammatory diseases and alcohol abuse were the exclusion criteria. We performed a full ophthalmologic examinationin all patients, which included best corrected visual acuity (BCVA), applanation tonometry, slit lamp examination, fundus examination. Lenticular opacity degree was evaluated according to LOCS III. To estimate the spontaneous oxidative modification of proteins to determine the level of protein carbonyl derivatives by the method of E.E.Dubinina. The level of modified proteins in the blood serum was measured on a spectrophotometer (at a wavelength of 370 nm) by the level of the formed 2,4-dinitrophenylhydrazones (2,4 -DNP), and calculated using the extinction coefficient and expresses as nmol/ml of blood plasma. Levels of sex steroid hormones in the blood was determined by immunoenzymatic analysis

Results:

Average index of Best-corrected visual acuity in SC group was 0,36 ± 0,04, and in СG it was 0,96 ± 0,02. Lenticular opacity degree in patients with senile cataract was 10,46±2,03. Found that males and females of reproductive age with age-related cataract, the initial level of testosterone and estradiol in the blood plasma was significantly decreased (p <0,05) by 16,5% and 15,8%, respectively, compared with the level of the test of the control group. It has been revealed that level of 2,4-DNP in blood plasma of patients with age-related cataract was on 192,2% significantly higher than in control group.

Conclusions:

Detected high levels of 2,4-DNP in in blood plasma of patients with age-related cataract, indicating severity of the process of oxidative modification of proteins in these patients. The results show that in patients of reproductive age with senile cataract high level of protein carbonyl derivatives are combined with low levels of sex steroid hormones in the blood serum FINANCIAL INTEREST: NONE
