Purpose:
To evaluate the influence of diabetes mellitus (DM) type 2 on total antioxidant capacity (TAC) of aqueous humor (AH) determined by copper reduction test, and the influence on antioxidant enzymes gene expression in lens epithelial cells obtained during the cataract surgery.
Setting:
Phacoemulsification induces inflammatory and oxidative changes in the anterior chamber (AC). Some reports suggest that DM can modify oxidative stress regulation by reducing capacity of antioxidant compensation of these changes. Control and diabetic patients without retinopathy were selected to harvest AH and lens epithelial cells to check these supposed changes
Methods:
Prospective, interventional, clinical study. Forty eyes from forty patients were included. They were divided into two groups: Group 1 (G1): twenty non-diabetic patients and group 2 (G2): twenty diabetic type 2 patients without retinopathy. Pure AH samples and lens capsules preserved in RNAlater, were harvested during surgery and preserved immediately at -80ÂșC until all the samples were collected. AH TAC was determined by copper reducing equivalents (CRE), and gene expression of catalase (CAT) and superoxide dismutase 2 (SOD2) were determined by real-time polymerase chain reaction (PCR) in anterior capsule lens epithelium. GAPDH was selected as housekeeper gene.
Results:
Mean values for TAC, measured by CRE (uM), were: G1: 5186,4 (SD = 2040,1) and G2: 5452,4 (SD=2225,9). No statistically significant differences were found among groups (p>0.05). CAT and SOD2 were expressed in both groups. Relative expression to GAPDH of CAT was 0,0020 for G1 and 0,0028 for G2; whereas relative expression to GAPDH of SOD2 was 0,0145 for G1 and 0,0227 for G2, no statistically significant differences were found between both groups (p>0.05).
Conclusions:
DM type 2 without retinopathy did not influence in AH TAC and relative expression of CAT and SOD2 genes.
