Corneal density quantification method using swept-source optical coherence tomography technology: comparison with Scheimpflug technology

Abstract Details

Purpose:

To establish the normal values of corneal density of healthy subjects using the anterior segment module of Swept source optical coherence tomographer (AS-SS-OCT) (Triton, Topcon, Japan) comparing to  Pentacam Scheimpflug system (Oculus, Inc., Wetzlar, Germany) and propose possible clinical applications of this values.

Hospital Clinic de Barcelona. University of Barcelona

Methods:

An AS-SS-OCT and Scheimpflug system were used to asses corneal density. The readings in healthy eyes were recorded by same technician after discard ocular comorbidities or prior ocular surgery. Two observers performed the analysis of the scans selecting the horizontal plane (0º-180º) and targeting the central mid-stroma. For AS-SS-OCT density was measured by using the online available image-processing ImageJ. Pentacam densitometry was measured using the Oculus density scale (0-100). Corneal thickness was also quantified by both instruments. The intraclass correlation coefficient (ICC) and repetitive coefficient (COR) were used to calculate the repeatability and reproducibility of measurements.

Results:

The mean densitometry value of normal corneas result in  16,26 for Pentacam and 112,33 for AS-SS-OCT. Pachymetry values were 547,95 and 524 respectively. The analysis ICC of densitometry values at central clear cornea suggested “excellent” reliability of more than 0.90 for both observers and both variables , and COR values were less than 0.30% for both variables within the same examiner. No correlation was found between the pachymetry and the densitometry values.

Conclusions:

Densitometry can be used as an objective measure of the corneal health and could be apply as a method for following up patients suffering from corneal dystrophies, corneal transplantation or post-infectious opacities.
