Experimental investigation of cAMP agonist effect on an in vivo corneal oedema model

Abstract Details

Purpose:

To evaluate the effect of a topically administered cAMP agonist on corneal oedema in vivo.

Setting:

Animal Laboratory for Medical/Scientific Purposes, ATTIKON Hospital, Athens

Methods:

Six New Zealand white rabbits (12 eyes) were involved. In Phase 1, corneal oedema was induced in all eyes by epithelium mechanical removal followed by instillation of Water For Injection drops over 30 minutes. In Phase 2, all right eyes were instilled with adenosine 3mg/mL at a constant rate of 4 drops/min (Adenosine Group) and all left eyes were instilled with NaCl 0,9% at the same rate (Control Group). Central Corneal Thickness (CCT) was measured with ultrasound pachymetry every 5 minutes. Percentage Thickness Index (PTI) and Thickness Change Rate (TCR) were calculated and analysed statistically with the indicated tests.

Results:

At the end of Phase 1, PTI increased significantly by +45,64%±0,35% (p<0,001), while TCR decreased significantly by -1,06%/min± 0,14%/min (p<0,05). In Phase 2, Adenosine Group showed significantly lower PTI and TCR values compared to Control Group at all time points (p<0,05).

Conclusions:

Topical administration of a cAMP agonist shows a significant retarding effect on corneal oedema in vivo.
