Effect of an antibacterial peptide for treatment of experimental infectious keratitis

Abstract Details

Purpose:

Bacterial keratitis is an ocular infection with the potential to cause significant visual impairment. Exploring of new antimicrobial agents for the treatment of antibiotic resistance keratitis is essential. In this study, we introduced a cationic peptide with antibacterial activity on methicillin resistance Staphylococcus aureus keratitis.

Setting:

An animal study conducted at Baqiyatallah University of Medical Sciences.

Methods:

Safety of cationic peptide was confirmed using the Draize scoring system previously. Efficacy of this peptide was evaluated by experimental infectious keratitis in the rabbit eye. After stromal injection of MRSA, the peptide was topically applied in the test group and compared with other groups including PBS, gentamicin, and synergy (gentamicin +peptide). After treatment, the eyes were clinically graded and the cornea was harvested for histopathology and colony counting

Results:

Colony forming unit in the peptide-treated group was significantly lower than gentamicin and PBS-treated groups. ( P< 0.05). Although the CFU for the synergy-treated group was lower than the peptide-treated group, this differences not significant (P>0.05). Histological feature indicated that structure of cornea was normal after administration of antimicrobial peptide

Conclusions:

Our results confirmed that the cationic peptide has significant potential for development as a new antibacterial agent for treatment of bacterial keratitis.
