Abstract Details
Purpose:
To evaluate the mean cell number (MCN), mean nuclear area (MNA), mean nuclear circularity (MNC) differences of human lens capsule epithelium cells (HLCE) between the pseudoexfoliation positive (PEX+) and negative (PEX-); trypan blue positive (TRYPAN+) and negative (TRYPAN-); extracapsular cataract extraction (ECCE) and phacoemulsification (PHACO) groups.
Setting:
The project is approved by Hacettepe University Medical Faculty Ethical Committee. the surgical process was performed in the department of ophthalmology and immunohistochemical studies were completed in the department of biophysics.
Methods:
History and demographic data of subjects were gathered.HLCE cells were obtained from anterior capsule material after cataract surgery. The nucleus of the cells in the capsule material was stained with 4′,6-diamidino-2-phenylindole(DAPI) stain and images were taken. ImageJ was used and a special code was written in order to binarize and watershed the images using artificial intelligence. By using this technique, the cell nucleus can be separated from each other and borders can be visualized. A 100µmx100µm square region was investigated in 5 random area and parameters were compared for each group using non-parametric tests for the analysis.
Results:
18 eyes of 17 subjects were enrolled in the study. Female to male ratio was 8/9 and mean age was 71.50±11.41. ECCE to PHACO ratio was 2/16. 5 cases have PEX+ and 3 cases underwent surgery with trypan blue. There was no significant difference between PEX+ and PEX- groups for MCN, MNA, and MNC (p=0.348, p=0.301, p=0.522). MCN was significantly lower in TRYPAN+ group compared to TRYPAN- (p=0.011), but there was no difference for MNA and MNC (p=0.314, p=0.767). MCN was significantly higher in PHACO group compared to ECCE(p=0.024) whereas MNA and MNC were not significantly different (p=0.574, 0.888).
Conclusions:
Image analysis methods are useful for morphological evaluation of the cells. Emerging technology allows us to better quantify the morphological features such as cell circularity. Trypan blue and ECCE could be said to have a decreasing effect on MCN of HCLE whereas this situation is not true for PEX+ subjects. Further investigations needed to reveal the underlying conditions.
