Abstract Details

Purpose:

This study aims to evaluate the in vitro effects of methylprednisolone, cyclosporine A and lysozyme on growth of Aspergillus sp. and Candida sp isolated from cases of infective keratitis.

Setting:

Tertiary care eye hospital in eastern India

Methods:

Aspergillus fumigatus and Candida albicans were grown in the presence of varying concentrations of methylprednisolone, cyclosporine A and lysozyme. Fungal growth was evaluated by measuring the optical density value in 96 well plates with RPMI 1640 medium and analyzed by fluorescence microscopy for biofilm determination. A semi quantitative measure of biofilm formation was calculated by XTT reduction assay.

Results:

Methylprednisolone had a statistically significant suppressive effect on the growth of A. fumigatus compared to lysozyme and cyclosporine while Candida albicans showed profuse growth in presence of steroids and no inhibitory effect was found for lysozyme and cyclosporine. However, biofilm formation was stimulated in the presence of steroids for both A.fumigatus and C.albicans while lysozyme and cyclosporine inhibited such formation for both.

Conclusions:

Biofilm formation of Aspergillus fumigatus and Candida albicans is stimulated by methylprednisolone and inhibited by lysozyme and cyclosporine.
