Abstract Details

Purpose:

To assess potential changes of the mechanical properties of the lens capsule following staining with brilliant blue (BB), indocyanine green (ICG) and trypan blue (VB).

Setting:

Experimental study, Department of Ophthalmology and Applied Physics and Center for NanoScience, Ludwig-Maximilians-University, Munich.

Methods:

Fifteen unstained lens capsules were obtained during uneventful cataract surgery. Sepcimens were dissected into seven wedge shaped parts. Three fragments were stained with 0.025% BB (Brilliant Peel®; Fluoron GmbH, Germany), 0.05% ICG (Pulsion, Germany) and 0.06% VB (vision blue ® DORC, Netherlands) respectively for one minute, another three specimens additionally illuminated using a standard light source. The seventh fragment served as an untreated control. All specimens were analysed using atomic force microscopy (AFM) in contact mode with a scan rate of 0.6 Hz. Two scan regions of 10×10?m were chosen and stiffness was determined by using AFM in a force spectroscopy mode. The force curves were performed with a data rate of 5000 Hz.

Results:

Staining of the samples resulted in an increase of tissue stiffness (BBG: p<0.001; ICG: p<0.01; VB: p<0.05). Additional illumination (indicated by "L") after staining did further increase the stiffness of the tissue but not significantly. Relative elasticity values were measured 1.61±0.15 for BB, 2.04±0.21 for BBL, 1.63±0.22 for ICG, 2.01±0.22 for ICGL, 1.23±0.11 for VB and 1.39±0.11 for VBL. In relation to unstained tissue the relative elasticity of the stained tissue increased by 1.2 fold following after illumination

Conclusions:

Staining significantly increases the mechanical properties of the human lens capsule
