Purpose:
To look at the effect of dilution of culture medium on lens epithelial cell growth

Methods:
We assessed the effect of dilution of culture media volume on LEC growth in a human donor eye model. Capsular bags from human donor eyes were maintained in either 1.5 or 6ml of serum-free culture medium which was removed and replenished every 48-72hrs for 28days. LEC growth was captured using phase-contrast microscopy and analysed using image J within the rhexis area. Culture media was retained for analysis of growth factors using Bioplex analysis and the capsular bags were bissected at 28days and retained for Bioplex growth factor analysis and immunocytochemistry.
Results:
LEC growth was retarded in the capsular bags cultured in 6mls media and morphological changes were seen in the pattern of cell growth. We postulate that this will be reflected by changes in the growth factor content.
Conclusions:
Increasing volume of culture medium will dilute the growth factor content and influence the dynamics of LEC growth.
