Trypan blue toxicity for cultured human trabecular meshwork cells

Purpose:

To investigate trypan blue’s (TB) at 0.06% concentration toxicity in cultured human trabecular meshwork (HTM) cells.

Setting:

Department of Ophthalmology, Friedrich-Alexander University Erlangen-Nuremberg, Erlangen, Germany

Methods:

Human trabecular meshwork (HTM) cells cultured in vitro were exposed to Trypan blue and acute toxicity was evaluated. Cells were exposed for 5, 30, 60 seconds and for 5,15,30,60 minutes to a commercially available TB preparation (Vision Blue; DORC International Zuidland, The Netherlands). Morphology was examined by phase-contrast microscopy and cell viability was measured using Hoechst 33342 and propidium iodide assays to resolve the proportion of living and dead cells.

Results:

Morphologic changes happened chiefly after 5 minutes of exposure to TB. Viability found to be 96.0%±3.6%, 94.8%±3.5% and 92.5%±4.4% after 5, 30 and 60 seconds of exposure respectively; a considerable toxic effect of TB was observed after 5,15,30 and 60 minutes of exposure with a viability of 85.0%±3.7%, 77.0%±6.7 %, 70.8%±5.9% and 68.3%±8.7% respectively.

Conclusions:

Trypan blue, in our model, proved to be not toxic, regarding cell viability, over an exposure time of up to 60 seconds; on the other hand further exposure results in an ongoing damage of cultured human trabecular meshwork cells. FINANCIAL DISCLOSURE?: No
