Experimental model of limbal stem cell deficiency in rabbits

Purpose:
This study was performed to achieve the model of total limbal stem cell deficiency (LSCD) in rabbits, and for a future use of this model for the corneal surface reconstruction using adipose-derived adult stem (ADAS) cells.
Setting:
Vissum Alicante, Spain.
Methods:
6 New Zealand albino rabbits were used to develop the model of severe unilateral LSCD. Visible corneal epithelium was removed using 100% ethanol. Superficial lamellar dissection of the limbal zone was performed 360º at an approximate depth of 30% of total limbal thickness. Chemical trauma on the central corneal epithelium with 100% ethanol was performed for 20 seconds. A 360º conjunctival peritomy was done. Impression cytology of the cornea and bulbar conjunctiva was performed before and after 2 months of surgery to confirm the LSCD. Examinations under slit-lamp were done at 1 and 2 weeks and 1 and 2 months.
Results:
All the animals presented 360º corneal neovascularization (NV) with affectation of the pupillar axis after the second month of the surgery. At this time we could observe central superficial corneal NV on half of them and central deep corneal NV on the other half. The corneal transparency decrease significantly, from moderate to completely opaque. The epithelial defect observed after surgery resolved in all the rabbits and no signs of hyperaemia or pain were found. Impression cytology of the corneal area at 2 months showed presence of conjunctival cells in all the eyes.
Conclusions:
This model of stem cell deficiency producing a 360º corneal neovascularization was successfully achieved and could be used for future alternative treatments for corneal surface reconstruction.
