The effect of sulfated glycosaminoglycans (sGAG) on human corneal epithelial cell proliferation

Abstract Details
Purpose:
To investigate the proliferation and cytotoxic effect of different concentration of sGAG on human corneal epithelial cells (HCECs)
Setting:
The S. Fyodorov Eye Microsurgery State Institution, Moscow, Russian Federation
Methods:
Corneal epithelial cell cultured in DMEM/F12 supplemented with 10% fetal bovine serum (FBS), HyClone III, L-glutamine (2,5 мМ) and antibiotics (penicillin steptomitsin ). In 1st group was exposed to 0,1% sGAG , in 2nd group –0,5% sGAG, in 3d group –1% sGAG, in control group- without treatment. For the real-time monitoring of cell growth and death we used the xCELLigence real-time cell analysis system . Proliferation effect was measured as the relative impedance change -cell index (CI), across microelectronic sensors integrated into the bottom side of the membrane
Results:
After 1 hour of treatment the CI values in 1st group (0,1% sGAG) reached of 1,33± 0,10 CI , In 2nd group (0,5% sGAG) 1,33± 0,16 CI , In 3d group (1% sGAG) 1,22± 0,13 CI , in control group- 1,55±0,02 CI.
After 2 hour of treatment the cell index (CI) values in 1st group reached of 2,38± 0,06 CI, In 2nd group 2,64± 0,26 CI, In 3d group- 2,55± 0,09 CI, in control group - 2,16±0,01 CI. After 3 hour of treatment the cell index (CI) values in 1st group (0,1% sGAG) reached of 2,54± 0,21 CI, In 2nd group (0,5% sGAG) 2,65± 0,24 CI, In 3d group (1% sGAG) 2,86 ± 0,11 CI, in control group- 2,37±0,02 CI
Conclusions:
In the groups cultured with sGAG at concentrations 0,1-1% cell index was significantly higher than that in the control group. sGAG in concentration 0,1-1% stimulates corneal epithelial cell proliferation
