In vitro cellular protective effect of FRpro OVD in free radical chemical and phaco energy damage simulations

Abstract Details
Purpose:
To investigate the cellular protection provided to tissue culture fibroblasts by a number of leading OVDs and FRpro under chemical free radical damage and phaco probe exposure conditions simulating phaco cataract surgery.
Setting:
In vitro cellular laboratory investigation, University of Brighton, UK, and York Finch Eye Associates, Toronto, Canada
Methods:
3t3 standard fibroblast cells (originally from Swiss albino mouse embryo) seeded at a cell density of 30,000 cells/cm2 and incubated for 24 hours were coated with 30-40µl (1mm depth) of leading OVDs. Following coating, 200 ml of Phosphate buffered saline (PBS) either supplemented with, or without, 0.75µM H2O2 was layered over the OVD’s. In those groups without H2O2, a Megatron Phaco probe tip was positioned just below the surface of the PBS, and run for 5 or 30 seconds at 60% power without irrigation or aspiration. Cellular viability was measured using MTS assay.
Results:
FRpro was found to provide significantly greater cellular viability following exposure than all other tested OVDs in both chemical and phaco cellular studies. A relationship between Phaco power and duration was found to significantly impact cellular viability in all treatment groups but with FRpro preforming better under all conditions.
Conclusions:
FRpro provides significantly greater protection against free radical & phaco induced cellular damage than the tested market leading OVDs. This in part may be due to its free radical scavenging high sorbitol content and/or it’s coating and barrier effect properties.
