Apoptosis of lens epithelial cells in patients with pseudoexfoliation syndrome

Abstract Details

Purpose:

The aim of this study was to measure the density of apoptotic lens epithelial cells (LECs) and to determine whether the effect to cataract formation in eyes with the Pseudoexfoliation Syndrome (PEX)

Setting:

Necmettin Erbakan University Meram Faculty of Medicine, Ophthalmology Department

Methods:

Twenty-one eyes of 21 patients with the diagnosis of PEX without glaucoma (Group 1), and 22 eyes of 22 subjects without PEX (Group 2) were enrolled in this study. During cataract surgery, anterior capsule samples were obtained by the 5.5 mm continuous curvilinear capsulorhexis method. Apoptosis was determined with a TUNEL kit (Boster, Wuhan, China) according to manufacturer’s protocol. The apoptosis rates were calculated by randomly choosing 4 fields in each stretched preparations under a microscope and 100 cells were counted in each field to calculate apoptosis rates.

Results:

Forty-three of 43 patients were enrolled. There was no significant difference in age and sex between the 2 studied groups (p>0.05). Under the a microscope, the LEC’s were stained light blue, their nuclei were oval shaped, positive stained cells were occasionally found in group 1. While in group 2, significant amount of brown and black-brown positive stained LEC’s appeared, their nuclei condensed. The apoptosis rates were 35.2±2.1% and 14.1±1% in Groups 1 and 2, respectively. The proportion of positive stained LEC’s were higher in group 1(p<0.001).

Conclusions:

The lens epithelial cell is one of the key mechanisms on lens structure maintaining, metabolism and function and also associated with cataractogenosis and its development. In this study, we compared LEC apoptosis rate among cataract patients with PEX and age related cataract. The results show that higher apoptosis levels in patient with PEX, which might be plays important role in the development and progression of cataract
