Purpose:
To investigate functions and expression profile of ANO/TMEM16 family in pterygium
Setting:
Yonsei University College of Medicine
Methods:
Pterygium tissues and circumferential normal conjunctiva tissues were obtained from the patients, who had pterygium surgery. Total RNA was extracted from the samples, which were acquired from the patients. The RNA samples were changed to cDNA, subsequently quantitative PCR was performed to analyze expression profile of ANO/TMEM16 family.
Results:
Among TMEM16/ANO family members, the expression of TMEM16C/ANO3 and TMEM16D/ANO4 were increased in pterygium tissues. TMEM16G/ANO7 expression was markedly decreased in pterygium tissues. Although the exact cellular function of ANO3, ANO4, and ANO7 is uncertain, ANO4 localizes in ER membrane and regulates intracellular calcium signals, and the expression of ANO7 reduced in high grade prostate cancer.
Conclusions:
We described expression profile of ANO family in pterygium. TMEM16A/ANO1 has been studied as a factor promoting proliferation in several cancer tissues. Although the expression profile of tumor associated genes in pterygium has been investigated in many studies, the tendency of ANO family in pterygium has not been studied yet. Among ANO family, ANO7, reduced in high grade prostate cancer, was also decreased in pterygium, which means ANO7 is associated with pterygium development. ANO4, regulator of calcium signaling which is important in cellular proliferation, also has a role in pterygium.
