Oxidative stress of crystalline lens in rat menopause model

Purpose:
Oxidative stress is a major cause of cataract formation. Our aim was to evaluate the lenticular oxidative stress by measuring total oxidant status (TOS), total anti-oxidant capacity (TAC) and oxidative stress index (OSI) of crystalline lenses in menopausal rat models.

Pamukkale University, Department of Ophthalmology and Physiology and Experimental Animals Unit, Denizli, Turkey
Methods:
Forty Wistar type female albino rats were included in this study. Thirty rats underwent oophorectomy to generate menopausal model and 10 rats formed the control group (Group 1). In the oophorectomy group, 10 rats formed the menopause control group (Group 2). One week after operation, 10 rats were administered daily methylprednisolone injection until the end of the study (Group 3). The remaining 10 rats were administered intraperitoneal streptozosin (50 mg/kg) to induce diabetes mellitus (Group 4). Eleven weeks after oophorectomy all rats were sacrificed and their lenses were extracted. TOS, TAC and OSI measurements of lenses were compared between groups.
Results:
The mean TOS value was the highest in group 2 and lowest in group 3. The mean TAC value was the highest in group 1 and lowest in group 3. The mean OSI was the lowest in group 1 and highest in group 4. The difference between the groups was not statistically significant in the aspect of OSI values (p>0.05)
Conclusions:
Our results indicate that menopause may promote cataract formation by increasing OSI. The co-existence of menopause and diabetes mellitus has a more prominent influence on increasing lenticular oxidative stress.
