Evaluation of corneal riboflavin penetration with two riboflavin solutions

Abstract Details

Purpose:

To evaluate riboflavin penetration ex vivo in human eyes using riboflavin solutions with dextran or hydroxpropyl methyl cellulose (HPMC).

Setting:

Medical University of Vienna, Department of Ophthalmology and Department of Rheumatology.

Methods:

Corneo-scleral buttons were prepared from human donor eyes not suitable for transplantation. After the epithelium was removed, riboflavin solution with dextran was installed on the denuded stroma for up to 20 mins (Group1). In Group 2, the corneas were soaked with a riboflavin solution with HPMC for up to 10 minutes. Riboflavin penetration was measured by calculating the transmission coefficient and by using two-photon confocal microscopy. In both groups at least 2 corneas were used for each measurement.

Results:

The Mean Fluorescence Intensity (MFI) increased with the duration of riboflavin application in Group 1 and showed a shift of the peak intensity towards deeper layers of the cornea with increased time. The same pattern of penetration was observed in Group 2. The MFI signal of corneas in Group 2 was lower compared to corneas in Group 1.

Conclusions:

In this ex vivo model, the measured Riboflavin concentrations were lower in eyes treated with a Riboflavin solution using HMPC instead of Dextran as a carrier vehicle.
