The effect of anterior stromal puncture using Q-switched Nd:YAG laser on corneal wound healing

Purpose:
This study aimed to evaluate the impact of normal healing and the anterior stromal puncture using Q-switched Nd:YAG laser treatment of injury on corneal protein after different periods.
Setting:
The energy setting of Nd: YAG laser was 0.5 mJ per pulse or less, the spot size was 10 microns; the cone angle was 16 degree; the wavelength was 1064 nm and the duration of the pulse was 4 nanosecond.
Methods:
21 rabbits classified into three groups. control group, The rest of the animals, corneal epithelium injured by needle and divided into:(A) Normal healing group: divided into three subgroups and the animals left for normal healing for1 day, 1 week, and 4 weeks, (B) Laser treated group: divided the same and subjected to anterior stromal puncture using Q-switched Nd: YAG laser and the animals were left for 1 day, 1 week, and 4 weeks respectively. the corneas were isolated for estimation of total protein content, sodium dodecyl sulfate polyacrylamide gel electrophoresis, total antioxidtive capacity, total oxidative capacity and oxidative stress index.
Results:
Corneal total protein showed increment in percentage change in normal healed groups after 1 day, 1 week, 4 weeks by values 93%, 68% and 39%. Nd: YAG laser treated group showed better improvement in corneal protein with percentage changes 58%, 29%, and 7.5%. In SDS- PAGE, a protein band at 110 KD appeared in epithelium for both normal healed and Nd:YAG laser treated group with changes in the peaks. after 4 weeks the peak at 110 KD disappeared in the injured epithelium treated with Nd:YAG. After four weeks, OSI in laser treated corneas showed balance between antioxidative and oxidative capacity.
Conclusions:
Conclusion: Anterior stromal puncture by Q-switched Nd:YAG laser is an effective, simple, safe and promising procedure to treat recurrent corneal erosion than normal healing.
