Purpose:
To evaluate the cytotoxicity of topical anti-inflammatory eye solutions on cultured human corneal epithelial cells in vitro.
Setting:
Medical Research Institute of Pusan National University Hospital.
Methods:
The cytotoxicity of topical anti-inflammatory eye solutions such as Bromfenac, Diclofenac, Pranoprofen, and Flumetholone was evaluated using the MTT assay according to 10, 50 and 100 fold dilution of the commercial eye drops. Also cellular damage was determined using the lactate dehyrogenase (LDH) assay after 1 hr, 4 hr, 12 hrs and 24 hours exposed to anti-inflammatory agents. During 48 hours, scratch test was determined using a migration and wound healing assay of corneal epithelial cells. Cellular morphologies were examined by performing transmission electron microscopy.
Results:
The metabolic activity of the corneal epithelial cells decreased and the LDH titer increased with more concentration of eye drops to Diclofenac and Flumetholone rather than Bromfenac and Pranoprofen. Especially, the LDH titers of 24 hours exposure at Flumetholone significantly increased rather than other anti-inflammatory agents. Cellular migration was not observed after exposure to Diclofenac and Flumetholone after 48 hours later. Severe cellular morphological damage was observed on transmission electron microscopy after exposure to Diclofenac and Flumetholone.
Conclusions:
Among several anti-inflammatory agents of topical eye solutions, Bromfenac and Pranoprofen induced less cellular damage to the cultured human corneal epithelial cells than did Diclofenac and Flumetholone.
