Abstract Details

Purpose:

Oxidative stress-induced DNA damage plays a critical role in the pathogenesis of age-related cataract (ARC) which has three subtypes including cortical, nuclear, subcapsular types. This study examined DNA methylation and histone modification of DNA repair gene WRN and their relationship with the gene expression in ARC patients.

Setting:

Eye Institute, Affiliated Hospital of Nantong University, 20 Xisi Road, Nantong, Jiangsu, China

Methods:

Western blot and qRT-PCR were used to analyze the expression of WRN. Methylation status of CpG islands within the WRN promoter were assessed by using Bisulfite-sequencing PCR (BSP). Chromatin immunoprecipitation (CHIP) was used to detect histone modification of the WRN gene.

Results:

The mRNA and protein levels of WRN were decreased in ARC cases. The CpG island of WRN promoter displayed hypermethylation in ARC cases. Acetylated H3-K9 was lower while methylated H3-K9 was increased in ARC cases.

Conclusions:

The evidence suggests that WRN expression was regulated by epigenetics in ARC.
