Enlightening intraocular lens calcification: an innovative in vitro model

Abstract Details
Purpose:
To develop a model suitable for practice to reproduce and investigate the process of intraocular lens (IOL) calcification.
Setting:
The David J Apple International Laboratory for Ocular Pathology, University of Heidelberg, Heidelberg, Germany.
Methods:
Calcification was reproduced using an electrophoresis and aqueous solutions of calcium chloride and disodium hydrogen phosphate adjusted to pH 7.4. The calcified IOLs were then analyzed using light microscopy, Alizarin red and von Kossa staining, scanning electron microscopy, energy dispersive x-ray spectroscopy, transmission electron microscopy and electron diffraction (ED). Obtained results were compared to published results of IOLs explanted and analyzed because of calcification.
Results:
Comparable with the calcified and subsequently explanted IOLs, Calcium phosphate deposits could be found within the polymer of every experimental IOL. ED analysis confirmed the presence of Hydroxyapatite in the IOL polymer, being the same type of calcium phosphate crystal in in vivo calcified IOLs. Comparison to previous published data about the calcium phosphate deposits of explanted IOLs confirmed similar results.
Conclusions:
The developed in vitro model can be used to reproduce and investigate IOL calcification.
