Purpose:
To evaluate ocular penetration of topically applied ceftaroline, a fifth generation cephalosporin, targeted against methicillin-resistant Staphylococcus aureus
Setting:
: Ophthalmology Clinic, Bolu State Hospital, Bolu, TURKEY
Methods:
Twenty New Zealand white rabbits were divided into 3 groups. One drop of 50 µl (8 mg/mL) ceftarolines was administrated in Group I (n=6). In Group II (n=6), eyes were dosed in accordance with a keratitis protocol (1 drop of 8 mg/mL every 15 minutes for 1 hour). Aqueous humor was sampled 5 times from immediately after to 2 hours after drop delivery. In Group III (n=8), a keratitis protocol was implemented before the animals were humanely killed. Conjunctiva, cornea, vitreous, and blood samples were collected 1 hour and 2 hours after the last drop in Group III. Ceftaroline concentrations were measured by high-performance liquid chromatography.
Results:
In Group I, the mean peak ceftaroline concentration in the aqueous humor (2.61 µg/ml) was 60 minutes after the last drop delivery. In Group II, the mean peak ceftaroline concentration in the aqueous humor (4.03 µg/ml) was 45 minutes after the last drop delivery. In group III, the mean concentrations of ceftaroline in the vitreous samples was significantly higher in the second hour after the last drop (7.05 µg/ml).
Conclusions:
Ceftaroline levels in the aqueous humor, and vitreous exceeded the minimum inhibitory concentration of most gram-positive organisms that cause bacterial keratitis and endophthalmitis. FINANCIAL INTEREST: NONE
